Roaches continued to incubate at 37 C for another 24 hours and survival will continue to be monitored.
Problem with Ppreps
I have been having some issues with Plasmid Preps recently. The Fisher lab received a new new Zyppy Plasmid Miniprep kit (Link) and we have been unable to successfully isolate plasmids. The neutralization buffer was not stored at refrigeration temperatures as the instructions suggest for about a month after the kit was received. However, after calling the manufacturer it was determined that there was plenty of RNase presence that the buffer should work fine still.
We have been unable to isolate the knockout vector pEx18Tc and the pTnMod-Okm plasmid using this new kit. Today, a previously created pEx18Tc::GW plasmid was brothed in TSB Tet Cm and incubated at 37 C shaking 225 rpm. I plan to attempt to isolate this plasmid to determine if there is a problem with the kit, or a problem with my pEx18Tc and pTnMod-Okm plasmids.
