The drop plate of the dilutions prepared of the B. thuringiensis spore stock from yesterday (incubated at 30C for 24 hours) were all completely overgrown. The spore stocks were not diluted down enough. Upon microscopic examination the stock appeared to be around 90% pure or higher. There are far more spores present than what is typically seen from the P. larvae spore preps.
The B. thuringiensis spore stock stored at 4C was further diluted down to 10^-8 in a ten fold serial dilution. The drop plate method was performed on dilutions 10^-5 to 10^-8 on LB only agar and incubated at 30C not inverted.
